LB, Terrific Broth, and S.O.C. medium are used for the maintenance and popagation of E.coli. Terrific supports higher cell densities than LB medium and is therefore, used for maximum plasmid yield. S.O.C medium is used as a recovery medium during the final stage of transformation.
An aseptic fill using a microbiological growth medium instead of product is a sensitive tool in determining the microbiological acceptability of a manufacturing process. This testing method enables all previous steps in the process to be assessed and, therefore, provides a realistic indication of microbial control of the process. Sterility validation uses tryptic soy broth (TSB) in place of the normal liquid product during the production process. If there is no indication of organism growth in the TSB following the production run, the process is considered an aseptic fill. TSB is also employed to test each final liquid product for sterility after each production run. TSB is inoculated with aliquots of the final product. If there is no indication of organism growth, the product is considered sterile. Mediatech offers TSB, also known as soybean casin digest, for use in sterility testing and validation. The product is a sterile ready-to-use solution for the simulation of aseptic filling of liquids and sterility testing. Due to the rich nutrient base, TSB may be used in the cultivation of fastidious microorganisms. This growth medium complies with current USP requirements.
